In 1942, the effectiveness of penicillin in experimental animal spirochaetosis was demonstrated by Lourie &Collier (1943). A year later, this antibiotic was introduced in the lreatmcnt of human syphilis (Mahoney ct al., 1943). In the following decade a complete reorientation of therapy took place not only in venereal sghilis of adults and in congenital syphilis, but also in the endemic treponematoses of childhood-yaws, pinta, and nonvenereal syphilis. Penicillin gradually came to snpplement-and subsequently to replace-the traditional therapy with arsenicals and/or bismuth. The marked treponemicidal effect of penicillin and the early low frequency of side reactions, as well as its ease of administration and low cost, made it appear the ideal drug. With the advent of such long-acting preparations as penicillin in beeswax and oil (POB) (Romansky & Rittman, 1945), followed by procaine penicillin G in oil with aluminium monostearate (PAM) (Buckwalter & Dickison, 1948) and benzathine penicillin G (DBED) (Szabo el al., 1951). Paul Ehrlich's dream of developing a single-dose injection treatment for treponematoses seemed to have been fulfilled. These developments were kept under international review in the context of the treponematoses programme of the World Health Organization (WHO). They have been periodically assessed by the WHO Expert Committees on Venereal Infections (World Health Organization, 1948, 1949; WHO Expert Committee on Venereal Infections, 1950a) and on Venereal Infections and Treponematoses (WHO Expert Committee on Venereal Infections and Treponematoses, 1953a, 1960a), a WHO Syphilis Study Commission in the USA (WHO Syphilis Study Commission, 1950b), two European symposia on therapy evaluation (World Health Organization, 1951a, 1951b), and a number of international congresses (British Medical Association, 1953; Hellerstrom et a]., 1959-60; Pillsbury & Livingood, 1963; Storck, 1964; US Department of Health, Education, & Welfare, 1964a; Spitzy & Haschek, 1967; Jadass o h & Schirren, 1968).
